The easiest way is to boot into the Finder, insert a blank disk into the
drive, and click "Yes" when Finder asks you if you want to format it.

Choose "ProDOS" or "HFS", 800K, and 2:1 interleave, and you're in business.

You do have System 6.0.1, don't you?  If not, things get a little trickier.
