In general, yes.

There may be a small number of exceptions, i.e. 8-bit applications which
use the Miscellaneous Toolset (so they will still be able to access the
mouse with slot 4 set to Your Card), or 16-bit applications which try to
use the mouse firmware (probably older applications only).
