how to access super serail card in langage machine?

i'm doing data acquisition and i don't want to use the command IN#2
for receiving data from the serial port.

i would prefer to receive data from a register or accumulator or a defined
address in machine langage.

Also i want to know how to initialise the card (always in machine language)
