No it doesn't. The root directory starts at block 2. It occupies four
adjacent blocks (for a normal volume), and is followed by the volume
bitmap, which varies in size according to the total number of blocks on
the volume (one bit per block, so divide the total number of blocks on
the disk by 4096 and round up to determine how many blocks are in the
volume bitmap). Blocks 0 and 1 are also used (boot code).
