That won't stop the 5.25" drive's select light blinking. As I said in my
earlier post, it is caused by the system polling the 3.5" drive, and is
a hardware issue with the way in which the drive select signals are
implemented.

Disabling the driver will prevent GS/OS trying to read the 5.25" drive
when it is searching for a disk by name or scanning all drives for the
next readable volume (which is horribly slow if there is a 5.25" drive
available).

If you will only very rarely need the 5.25" drive but don't want to
unplug it, you can also set Slot 6 to "Your Card" in the Control Panel,
which will prevent the 5.25" drive being available anywhere unless you
change the setting back and restart.
