It depends on the software using the card.

If the software depends on the on-card firmware interface, then the
slot must be set to your card.  If it uses the card's hardware directly,
as if it were, for example, an Apple Parallel card, then the firmware
on the card is not used and the slot can be set either way.

Any use that must be set up by doing a "PR#s" command, like
printing from Applesoft, relies on the on-card firmware, and will
require the slot to be set accordingly.

On the other hand, any software that includes its own hardware
driver, for example, to print NLQ multi-font output, bypasses
the on-card firmware, rendering slot setting irrelevant.
